Optimization Studies on Imatinib Mesylate Loaded Nanoliposomes Using Box-Behnken Design

Nanoliposomes are bilayer phospholipid vesicles used to encapsulate and deliver therapeutic agents. The study was aimed investigate the effects of critical variables on nanoliposomes characteristics. Imatinib mesylate-loaded were formulated by two-step emulsification process using a high-speed homogenizer system probe-type ultrasonicator. Box-Behnken design utilized optimize parameters. mean particle size found be 211 nm 623.3 with low value polydispersity index (0.005 0.7). Zeta potential values varied from ‒27.6 mV ‒9.2 in uncoated +27.5 chitosan-coated nanoliposomes. encapsulation efficiency formulation NLP-H8 containing 200 mg phosphatidylcholine, homogenization speed 12000 rpm, 7 min sonication time 76.49% without coating 85.4% 0.2% w/v TEM image confirmed spherical shape In-vitro drug release demonstrated that optimized nanoliposomal formulations released 84.67% loaded after 24 h 0.1 N HCl. IC50 7.98 μM. Nanoliposomal prepared successfully suitable size, morphology, efficiency, release. models developed this may further as response surface for various parameters
